To do the mission to unlock her, first you would have to have finished the mission Severing the Roots. After doing this, you can go to the market to buy (for 175 tokens) the scroll:
"Vines of Betrayal"

This unlocks the mission "A Missing War Dancer" which, upon completion will unlock another mission, which unlocks another, and so on and so forth until you get to the final mission.
Mission 1: A Missing War Dancer
• Win 3 battles in a row
• Win 1 battle with Kusa Niwa
• Win 1 battle with Kusa Kao
• 12 Collaboration Points (Minus 3)
This dialogue begins in the Kusa Village.
: (Insert Username)! Something's wrong! Yugana's been missing for 5 days! We think she's been abducted! Please (Insert Username), help us!
: Niwa, calm down, everything will be fine. That being said, would you be willing to help us, (Insert Username)?
: If it's ok with you, captain, then I think we should. Niwa, Kao, you said she's been missing for 5 days. What did she leave for in the first place?
: She left on a mission to pick some herbs for our village's medics. This task normally would've taken her under an hour. She's yet to return though. I really can't think of any possible explanation.
: Exactly where did she go to pick those herbs, Kao? I mean, did you search the area where she should've been?
: Yes, we've sent a group of clan members to search the area. No success however. This whole situation feels fishy to me.
: Ok, so we know she's not in the area where she was supposed to be picking the herbs. If that's the case though, what exactly would you like you us to do about it?
: Normally, I'd think that extra help would prove to be a waste, and a setback. Yugana's too important to our clan's offensive line to lose though, and right now, I'm desperate for extra eyes to search for her. What I'm asking is that you'll lend you and your colleagues' attention toward searching for her. Assuming you're willing to help, please hurry!

: Kao, don't make me repeat myself. When I left, I left the village not as a mission, but because I wanted to leave. I have no plans of coming back. Please, again, leave, and forget we even had this encounter.
: Yugana! What's going on? What's wrong with you? Can you even hear what you're saying right now? Seriously, just come back with us!
: Niwa, I'm not coming. I don't have time to waste arguing though, so I'll tell you exactly what's going on. I've betrayed the Kusa, by assisting Jikkousu in faking her death. Now, before things become unpleasant, I'll be leaving.
: YUGANA!!
: Kao, what do we do now? I mean, do we follow her?
: No. Although it pains me to think of Yugana as an enemy and traitor, that's what she is. As of right now, our current objective is to capture her, or if that's not possible, kill her.

: You're quite arrogant. That being said, you're quite right. In addition to my power however, Yugana will fight against you too.
: I myself could defeat Yugana without anyone else's help. How do you think we'll lose, even with her fighting us?
: Ha! While you may have been able to defeat me back then, I now harbor more power than you can imagine, all thanks to Jikkousu. Yes, now you get it; I have an aging crystal, which with my war-dancing, can easily crush you, Kao.
: Only one way to know for sure... Attack!

Yugana has left the Kusa, after deciding to help Jikkousu. She's become ever so much stronger due to Jikkousu's help, and now hosts more power than ever before.

Snarling Vines
Yugana combines her plant manipulation with Jikkousu's aging crystal given to her.
Yugana deals 10 affliction damage to one enemy. If they use a skill next turn, their cooldowns will be increased by 2 for 2 turns.* If they don't use a skill next turn, the target will take another 10 affliction damage.
*Only one increase can be active at a time. If used while a cooldown increase is in effect, they will take 20 affliction damage instantly.

Blossoming Desperation
Yugana uses her enemies' pain, and converts it into life for her allies.
For 3 turns, each time an enemy takes 20 or more damage, Yugana and her allies will heal for 5 health.

*If Yugana is stunned during use, it will have no effect that turn. When the stun ends, however, the skill will take effect again.

Dance of the Petals
Yugana scatters her body into a horde of petals, confusing her enemies.
For 1 turn, each enemy that uses a skill will be stunned for 1 turn. This is invisible.

Peace of Mind
Yugana turns the remains of her energy into a flower which brings peace to her.
This skill makes Betraying Yugana invulnerable for 1 turn.

What I'm going to do for this is post the previous ways she's been.
v.1:
